Bu çalışmanın amacı endemik, nesli kritik tehlikede olan ve tıbbi Micromeria dolichodontha P.H. Davis türünün in vitro aksiller sürgün yoluyla çoğaltma protokolünü belirlemektir. In vitro şartlar altında tohumların çimlendirilmesi ile elde edilen bitkicikler aksiller sürgün çoğaltımı için eksplant olarak kullanılmışlardır. Bitkicikler köklerinden ayrılarak farklı tip ve konsantrasyonda sitokinin (BA, KIN veya TDZ) içeren ve kontrol olarak herhangi bir bitki büyüme düzenleyicisi içermeyen Murashige & Skoog (MS) besin ortamlarında kültüre alınmışlardır. En yüksek sürgün sayısı 0.1 mgL-1 benziladenine (BA) içeren MS ortamında ve en uzun sürgünler 0.01 mgL-1 BA içeren MS ortamında oluşmuştur. Sürgünlerin köklendirilmesi için farklı tip ve konsantrasyonda oksin (IBA veya IAA) içeren ve herhangi bir bitki büyüme düzenleyicisi içermeyen ½ MS ortamı kullanılmıştır. En yüksek kök oluşumu (% 75) 0.5 mgL-1 IBA içeren ½ MS ortamında gözlenmiştir.


The goal of this paper was to determine the protocol for in vitro axillary shoot propagation of Micromeria dolichodontha P.H. Davis, an endemic endangered and medicinal plant. Plantlets obtained from in vitro germinated seeds were used as explants for axillary shoot propagation. The plantlets were separated from their roots and cultured on Murashige & Skoog (MS) media wihout and with different types and concentrations of cytokinins (BA, KIN or TDZ). The maximum number of shoots was formed on MS medium with 0.1 mgL-1  benzyladenine (BA), and the highest seedling length was formed on MS medium with 0.01 mgL-1 BA. ½ MS medium wihout and with different types and concentrations of auxin (IBA or IAA) were used for rooting of shoots. The maximum root formation (75 %) were observed on ½ MS medium with 0.5 mgL-1 IBA.
